9-Hydroxybenzopyrene is a reactive, dna binding compound. It binds to nuclear DNA and alters the transcription process. 9-Hydroxypyrene has been shown to be a potent mutagen at high doses in yeast (Saccharomyces cerevisiae) and rat liver microsomes. Low bioavailability of 9-hydroxypyrene is due to its rapid metabolism by human liver enzymes, which convert it into metabolites, such as 1'-hydroxybenzopyrene and 3'-hydroxybenzopyrene. These metabolites are also found in urine samples and have been shown to bind to cellular receptors such as epidermal growth factor receptor, leading to DNA synthesis and cell proliferation. 9-Hydroxypyrene has been shown to cause tumorigenicity in primary keratinocytes, hamster v79 cells, and model systems.
