Mestranol is a synthetic estrogen that is used in oral contraceptives. It is a prodrug of ethinyl estradiol and is metabolized to it by esters, conjugates, and glucuronides. The 2-hydroxy mestranol metabolite has been shown to be excreted in the bile, where it undergoes enzymic hydrolysis to form ethinyl estradiol. This drug has a high degree of soluble protein binding and has an irreversible binding affinity for microsomal proteins. Mestranol also irreversibly binds to enzymes such as cytochrome P450 and glutathione S-transferase.
