N-Hydroxymaleimide is a reactive amide that is used as an oxidation catalyst in the synthesis of monoclonal antibodies. It is also used in the preparation of conjugates for immunological assays. N-Hydroxymaleimide is typically synthesized by liquid phase methods, which are more efficient than solid phase methods. The reaction proceeds through the dehydration of an amine with a primary or secondary alcohol to form an amide. This process is irreversible and must be carried out under mild conditions to avoid side reactions with functional groups on the reactants. N-Hydroxymaleimide can be purified by gel chromatography methods before use, but it cannot be stored for long periods because it reacts with water, trifluoroacetic acid, and many other substances.
