5-Iodo-7H-pyrrolo[2,3-d]pyrimidine is a palladium-catalyzed cross-coupling reaction that can be used to synthesize 5-(iodomethyl)benzofuran. The palladium catalyst is generated in situ from the reaction of palladium chloride and triethylamine. The reaction uses a Suzuki coupling with 2 molar equivalents of 7H-pyrrolo[2,3-d]pyrimidine and 1 equivalent of phenylboronic acid. It proceeds via an oxidative addition followed by reductive elimination. The product is purified by column chromatography using silica gel as the stationary phase.
