5-Iodo-2-pyrimidinone-2'-deoxyribose is a radiosensitizer that has been shown to be effective against cancer cells in vivo. This compound binds to nuclear DNA and enhances the sensitivity of cancer cells to ionizing radiation. The oral prodrug 5-iodo-2,4(1H,3H)-pyrimidinedione (5IPD) is metabolized in vivo by esterases into 5-iodo-2,4(1H,3H)-pyrimidinedione 5'-phosphate (5IPDP), which is then converted into 5-iodo-2,4(1H,3H)-pyrimidinedione 2'-deoxyribose (5IDPR). 5IDPR is incorporated into DNA and prevents replication of the human telomeres. This compound has not been shown to have toxic effects on human liver or blood cells.
