Imatinib-d8 is a new analytical method that can be used to measure the concentration of imatinib in human serum. It is based on LC-MS/MS and uses a stable isotope-labelled internal standard (d8-Imatinib).
The concentration of imatinib in blood plasma has been shown to be proportional to the amount of drug administered, with an elimination half-life of about 2 hours. Imatinib binds to the BCR-ABL protein kinase, which leads to inhibition of its activity. This inhibition limits the production and release of hematopoietic cells from bone marrow, including stem cells and progenitor cells. The drug also inhibits growth factor receptors on stromal tumours.
