- Daridorexant
Nemorexant is an analog of the natural hormone melatonin, which is a circadian rhythm regulator. It has been shown to be effective in treating insomnia and sleep disorders by promoting sleep during the night and reducing daytime drowsiness. Nemorexant has demonstrated dose-dependent effects, with higher doses producing more complete suppression of the wakefulness-promoting system. The pharmacokinetics of nemorexant have been studied in healthy volunteers, cirrhotic patients, and in patients with hepatic impairment. It has a long elimination half-life (about 87 hours) and is eliminated mainly by hepatic metabolism, resulting in lower plasma concentrations for prolonged periods after administration. Nemorexant can also cause drug interactions because it induces CYP1A2, CYP3A4, and CYP2D6 activity.
