Leupeptin is an enzyme inhibitor that is used to study nicotinic acetylcholine receptors. It has been shown to be reactive in plasma glucose assays and to have a phenotype that is similar to that of type 2 diabetes mellitus in animals. Leupeptin inhibits the activity of viruses such as herpes simplex virus and pseudorabies virus by binding to the nicotinic acetylcholine receptor on the surface of cells. Leupeptin also inhibits autophagy, which may be due to its ability to inhibit the synthesis of proteins necessary for this process. The drug has been shown to act as a morphogenetic protein and trigger differentiation of bovine embryonic stem cells into cardiomyocytes. Leupeptin also binds with high affinity to the hydrophobic pocket on protein kinase C-α (PKCα) and blocks its activation, leading to apoptosis or cell death.
