5-Methylbenzimidazole is an isomer of benzimidazole that has been shown to have antimycobacterial activity. It inhibits the growth of Mycobacterium tuberculosis and Mycobacterium avium by binding to the corrinoid cofactor, which is essential for the formation of mycobacterial cell walls. 5-Methylbenzimidazole has been shown to inhibit the synthesis of vitamin B12, which may be responsible for its effects on mycobacteria. Its mechanism of action is not well understood but it may involve hydrogen bonding or vibrational interactions with water molecules. 5-Methylbenzimidazole was first synthesized in 1925 and its X-ray crystal structure was determined in 1960.
