Synthetic strategies for the synthesis of Methyl 2-amino-5-benzyl-1,3-thiazole-4-carboxylate (MTBA) have been developed and validated. MTBA is a synthetically accessible antibiotic with potent activity against mycobacteria. It targets the synthase enzyme that is involved in the synthesis of mycolic acids, which are essential components of the cell wall. The template strategy involves the use of a natural product as a template to obtain analogs with new properties. This strategy has been successfully used to develop MTBA derivatives. The synthesized compounds were tested against Mycobacterium tuberculosis and Mycobacterium avium isolates resistant to other antibiotics.
