3-Methyl-3H-diazirin-3-ol is a potent neurotoxin that inhibits the synthesis of proteins involved in the regulation of neuronal functions. It has been shown to inhibit synuclein, which is associated with neuropathological conditions, and can be used to induce Parkinson's disease in animals. 3-Methyl-3H-diazirin-3-ol binds to alpha synuclein and inhibits its expression. The binding of this compound to alpha synuclein has been shown using titration calorimetry, fluorescence resonance energy transfer, and caco2 cells. 3-Methyl-3H-diazirin-3-ol also binds to the regulatory domain of alpha synuclein, inhibiting its phosphorylation by protein kinase A and protein kinase C. This inhibition prevents the activation of transcription factors such as CREB and NFAT.
