Methyl thiocarbamate sulfone (MTES) is a reversible inhibitor of mitochondrial aldehyde dehydrogenase. It is used as an irreversible inhibitor in order to study the kinetics and cellular location of this enzyme. MTES irreversibly binds to the mitochondrial matrix and endoplasmic reticulum, inhibiting cellular respiration by preventing the conversion of acetaldehyde to acetic acid. This agent also inhibits glutathione reductase activity, which prevents the regeneration of reduced glutathione from oxidized glutathione. MTES has been shown to inhibit mitochondrial dehydrogenases, which are enzymes that catalyze the oxidation of substrates for ATP production. MTES is also known to target other enzymes, including alcohol dehydrogenase and cytochrome P450s.
