1-Methyl-nicotinamide methosulphate (1MNMS) is a drug that can be used in the treatment of nicotine addiction. It is an iminium chloride that binds to nicotine, forming nicotinamide and 1-methylpyridinium ion. This binding prevents the metabolism of nicotine by the liver, which leads to a decrease in the production of cotinine and other metabolites. The affinity of 1MNMS for nicotine is approximately 2 times higher than for nicotinamide, and it has been shown that this drug has a metabolic transformation mediated by CYP2A6, CYP2B6, CYP2C8 and CYP3A4 enzymes. In addition, 1MNMS can react with oxygen molecules to form an aldehyde intermediate or an aldehyde oxidase (ALOX) metabolite.
