4-Methoxyindole-2-carboxylic acid is a potent inhibitor of 3clpro, an enzyme that catalyzes the cleavage of peptide bonds. The affinity and inhibitory potency of 4-methoxyindole-2-carboxylic acid were determined by isothermal titration calorimetry and potent inhibition was observed at nanomolar concentrations. The dipeptidic modification led to increased inhibitory potency, while modifications in the methoxy group decreased this activity. Optimization studies have been carried out to increase the potency of 4-methoxyindole-2-carboxylic acid as an inhibitor of 3clpro.
