Met-enkephalin is a potent opioid that binds to mu, delta and kappa receptors in the brain. It is synthesized from Phe-enkephalin by removal of the C-terminal amino acid Phe. Met-enkephalin is a peptide that can be reversibly converted between two forms, A and B, by removing and adding a hydrogen ion. The A form has an active carboxyl group while the B form has an inactive carboxyl group. Met-enkephalin is synthesized by solid phase synthesis on a resin that is regenerated in neutral ph buffer. This drug has been shown to inhibit ATP levels in Caco-2 cells and decrease dopamine release in vitro. It also inhibits receptor binding experiments on rat brains and reversibly inhibits the binding of enkephalins to their receptors at different pH values.
