Neurotensin (NT) is a peptide that has been implicated in the regulation of feeding behavior, cardiovascular function, and pain perception. The neurotensin receptor is a G-protein coupled receptor with seven transmembrane domains. Neurotensin binds to the receptor and activates it by inducing intracellular calcium mobilization and neurotransmitter release. This activation can be dose-dependent, as seen in experiments using rat brain slices incubated with different concentrations of NT. Neurotensin also induces maximal activation at low concentrations, which may be due to its ability to bind to both extracellular and intracellular receptors. NT binds selectively to ventral tegmental area neurons from rats, leading to increased spontaneous firing rates. Cancer cells have been shown to express high levels of neurotensin receptors on their surface membrane, which may contribute to tumorigenesis and metastasis.
