Naphthol AS-BI-phosphate is a substrate for the enzyme acid phosphatase. It is used as a histochemical stain in human serum, which can be applied to detect the presence of alkaline phosphatase in tissue. Naphthol AS-BI-phosphate also has protease activity and inhibits the enzyme activity of diazonium salt. It is a substrate for p-nitrophenyl phosphate and undergoes oxidation, forming an acid product that can be detected by acid phosphatase. This product reacts with nitroblue tetrazolium (NBT) to produce a blue precipitate, which can be observed with a microscope or under UV light. The optimum pH for this reaction is 7.5 and it is inhibited by disulfide bonds.
