Nitrofluorescein, isomer 2 (NIF-2), is a fluorescent compound that has been used for the diagnosis of a number of diseases. It can be synthesized by reacting methanesulphonic acid with diacetate, which produces NIF-2. NIF-2 has been shown to be useful in the diagnosis of amyloidosis and other diseases where there are high levels of nitrosoamines in the body. This compound is also used to study the distribution of substances in biological tissues by filtering out the fluorescence that it emits. The fluorescence from this compound can also be quenched when it interacts with orthophosphoric acid or dipropionate. Nitrofluorescein, isomer 2 can also be hydrolyzed by a variety of enzymes such as esterases or glucuronidases, resulting in its active form, nitrofluorenone, which can then react with aminofluorescein
