6-Nitro-2-Piperazin-1-Yl-Quinoline is a potent uptake inhibitor of dopamine. It has been shown to increase the concentrations of 5HT in rat striatum, and it also inhibits locomotor activity in mice. 6NPQ binds to the dopamine transporter, which prevents the reuptake of dopamine by cells. This binding also results in an increased release of dopamine from nerve endings. 6NPQ has been shown to have a cytotoxic effect on carcinoma cell lines and is used as a model system for studying uptake inhibition. 6NPQ has been shown to bind to the amino acid methiothepin and other similar compounds, which are taken up by cells through the same transporter that 6NPQ blocks.
6NPQ is synthesized through a novel asymmetric synthesis method that involves reacting piperazine with nitroethane (nitro group) followed by reaction with hydrochloride acid (HCl).
