Nitro blue tetrazolium diformazan (NBT) is a dye that is used to measure the viability of cells. It can be used to detect cancerous cells in culture, as well as to monitor the growth of tumor xenografts in mice. The NBT assay can be performed using either K562 human leukemia cells or murine leukemia L1210 cells. In order for the NBT assay to work, the cells must be incubated with a tetrazolium chloride solution and then exposed to radiation. Cells that are still viable will take up the NBT dye and turn blue due to reduced nitro groups. However, this assay cannot distinguish between living and dead cells because all dying cells will appear blue. A control sample containing no radiation is needed in order to determine sensitivity of this assay, which is typically around 20%.
Nitro blue tetrazolium diformazan can also be used in conjunction with fatty acids such as oleic acid
