Hydrocortisone is a corticosteroid that has been shown to have anticancer activity. Hydrocortisone inhibits the growth of l1210 murine leukemia cells by inducing cell differentiation and arresting the cell cycle at G2/M. Hydrocortisone also induces apoptosis in leukemia cells, which may be related to its ability to inhibit lipid synthesis and activate protein kinase C. Hydrocortisone is metabolized into 2-oxa-3-aza-bicyclo[2.2.2]octane, a nontoxic metabolite that has no anticancer activity. It is not clear how hydrocortisone exerts its antitumor effect, but it may be due to an increase in the production of reactive oxygen species or inhibition of DNA synthesis or function.
