JWH-018 is a synthetic cannabinoid receptor agonist with a strong affinity for the CB1 and CB2 receptors (EC50 = 102 nM and 133 nM, respectively)1. Binding to the receptors triggers a reduction of intracellular cAMP levels by inhibition of adenylate cyclase. In vivo studies on experimental mouse models showed similar responses as THC, including hypothermia, catalepsy, and hypolocomotor effects2. In vitro, JWH-018 impaired electrically-induced synaptic transmission as well as LTP and glutamate/GABA release in ex vivo hippocampal slices. All behavioural and neurochemical effects could be prevented by pre-treatment with a CB1 receptor antagonist3.
