The lipolytic enzymes (or esterases) are a group of proteases that hydrolyze esters. They are found in the pancreas, where they are important for the digestion of fat and other lipids. The lipolytic enzymes catalyze reactions by using a proton, amide, or serine residue as nucleophile to attack the carbon-carbon bond of the fatty acid ester. It is thought that these enzymes bind to the substrate through hydrogen bonding with an amide or serine residue. Z-D-phenylalanine 4-nitrophenyl ester is an inhibitor of these enzymes and has been shown to reduce lipogenesis in laboratory mice by inhibiting pancreatic beta-oxidation of fatty acids.
