Sulforhodamine 101 is a fluorescent probe that has been used as a model system to study the effects of apoptotic proteins on cell death. It binds to nuclear DNA and fluoresces when excited with light at 480 nm, which can be detected using a fluorimeter. Sulforhodamine 101 has been shown to have pro-apoptotic properties in cells from human HL-60 leukemia cells and mouse brains. In these experiments, sulforhodamine 101 caused the release of calcium ions from intracellular stores by binding to the ryanodine receptor (RyR1) and inhibiting its function. This led to neuronal death by activating caspases, which are proteases that cleave other proteins. Sulforhodamine 101 also inhibited P-glycoprotein (Pgp) activity in brain tissue samples from rat models, suggesting it may be useful as an inhibitor for this protein in pharmacological agents for brain functions such as memory or learning.
