O-Tolylisocyanate is a potent antagonist of the enzyme dihydrofolate reductase, which is required for the synthesis of DNA. It also inhibits other enzymes involved in nucleotide synthesis, such as ribonucleotide reductase and thymidylate synthase. O-Tolylisocyanate has been shown to be effective for the treatment of inflammatory diseases, such as rheumatoid arthritis, Crohn's disease and ulcerative colitis. Additionally, it has been shown to inhibit the production of amyloid beta protein in Alzheimer's disease patients. O-Tolylisocyanate binds to human serum albumin and forms an insoluble polymer that prevents the release of hydrogen peroxide from neutrophils into tissues. This decreases inflammation by decreasing oxidative stress on tissues.
