Tritylvalsartan is a prodrug that is metabolized to valsartan in the liver. It is an angiotensin receptor blocker (ARB) used for the treatment of chronic liver disease, heart failure and hypertension. Tritylvalsartan is a racemic mixture of two stereoisomers, cilexetil and benzonitrile, which are formed by the Suzuki coupling reaction with a chiral auxiliary. The drug has been shown to be effective in treating cardiovascular diseases with no serious side effects. Tritylvalsartan forms an ion-pair with chloride in the blood plasma and inhibits angiotensin II-induced constriction of blood vessels. It also acts as a competitive antagonist of the angiotensin receptors AT1 and AT2, blocking their activation by angiotensin II.
