Thiamine thiazolonepyrophosphate (TTPP) is a coenzyme of the enzyme pyruvate dehydrogenase and is involved in the conversion of pyruvate to acetyl-CoA. TTPP binds to mitochondria, where it inhibits the activity of proton pumps and prevents cell lysis. It also has been shown to have inhibitory activities against enzyme form and holoenzyme, as well as substrate binding. TTPP forms an analog with thiamine (thiamin diphosphate), which inhibits spermatozoa motility by binding to the lipid-rich region of the plasma membrane. This coenzyme also has a role in lipoprotein metabolism, where it helps cells produce high density lipoproteins (HDL). TTPP is synthesized from thiamine mononitrate and two molecules of pyrophosphoric acid.
