Tiospirone is a drug that has been approved in Japan and South Korea for the treatment of depression. It is a serotonin-receptor antagonist, acting primarily as a 5-HT1A receptor partial agonist. Tiospirone is also an antagonist at 5-HT2 receptors. The drug binds to the acetylcholine, dopamine, and GABA receptors with low affinity.
Tiospirone has been shown to be effective in clinical trials for long-term efficacy and to be safe in therapy groups with or without concomitant use of psychotropic drugs such as ziprasidone.
