Ticlopidine N-oxide is a metabolite of ticlopidine, an anti-platelet drug. Ticlopidine N-oxide has been shown to be a competitive inhibitor of cytochrome P450 enzymes. The molecule is linear and the active site contains chlorophenyl groups that are oriented in a way that allows for the formation of the n-oxide form. X-ray crystallography studies have shown that ticlopidine N-oxide binds to the thiol group of cytochrome P450 enzymes, forming a covalent bond. This inhibits protein synthesis by preventing the transfer of electrons from NADH to cytochrome P450, which is required for metabolism. Ticlopidine N-oxide also forms dimers with other molecules, which may be responsible for its anti-inflammatory properties.
