Guaiacol is a phenolic compound that has been used as an antioxidant and to inhibit the formation of melanin. Guaiacol is also a precursor to the sesquiterpene lactone, guaiaretic acid. The guaiaretic acid has been shown to have bone growth-promoting effects in vitro and in vivo, inhibiting the activity of enzymes involved in melanogenesis. Guaiacol can be synthesized from catechol by oxidation with hydrogen peroxide, or via the hydrolysis of glycerol with sodium hydroxide. The guaiacol synthesis reaction products are fatty acids such as pentadecanoic acid and heptadecanoic acid. The enzyme preparations that catalyze this reaction are called lipoxygenases (LOX). Lipoxygenase activation energies range from -3.2 to -2.7 kcal/mol for different LOX enzymes, which may account for their different rates of activity.
