Trimethyl orthobenzoate is a chemical that is the product of the acylation reaction between trimethyl orthoformate and benzoic acid. The synthesis method for this compound was discovered in 1891 by Hermann Kolbe, who found that it could be prepared by reacting benzoic acid with formaldehyde and potassium hydroxide. Trimethyl orthobenzoate has been used as a precursor for the synthesis of other chemicals, such as nintedanib (used to treat liver cancer). Nintedanib inhibits the enzyme DPP-IV, which is involved in the production of proteins that are important for cell growth. Trimethyl orthobenzoate also has an anti-inflammatory effect due to its ability to inhibit protein kinase C.
