Tri(m-tolyl)phosphine is an organophosphorus compound that inhibits the polymerization of polyvinyl chloride. It has been shown to inhibit the growth of fibroblast cells in vitro and has a cytotoxic effect on human adenocarcinoma cells. Tri(m-tolyl)phosphine binds to the active site of polymerase, thereby inhibiting DNA synthesis by preventing incorporation of nucleotide monomers into the growing chain. The binding constants for tri(m-tolyl)phosphine to calf-thymus DNA and human adenocarcinoma cell DNA are 1.7 x 10 M and 1.4 x 10 M, respectively. This molecule is also a potent inhibitor of heterocyclic amines, which have been implicated as cancer-causing agents in cooked meat products such as bacon and hot dogs.
