Voclosporin is a drug that belongs to the class of immunosuppressants and is used to treat autoimmune diseases. Voclosporin inhibits P-glycoprotein (P-gp) and blocks the transport of some drugs from the gut into the bloodstream. The effect of voclosporin on P-gp has been shown using in vitro studies, x-ray crystal structures, and in vivo animal models. Voclosporin also inhibits inflammatory substances such as prostaglandins and leukotrienes, which are produced by immune cells and contribute to inflammation. This drug has also been shown to be effective against eye disorders such as uveitis, retinal detachment, and macular edema. Voclosporin is metabolized through a number of metabolic reactions including CYP3A4/5 enzyme oxidation or glucuronidation followed by a number of other reactions including cleavage by carboxylesterase or esterases to form 5′-desacet
