Description:

INI-43 is a small molecule that is a potent inducer of apoptosis. It binds to the TNF-related apoptosis-inducing protein (TNFAIP) and induces apoptosis in cancer cells by activating caspases. INI-43 also inhibits the nuclear translocation of NF-κB, which is a transcription factor that regulates proinflammatory cytokines. This inhibition prevents inflammation and tumor growth. The binding of INI-43 to TNFAIP leads to a conformational change in TNFAIP that activates caspases, resulting in apoptosis. In vitro assays have shown that INI-43 has an IC50 value of 0.1 nM for cervical cancer cells and 1 nM for mammary adenocarcinoma cells. The binding affinity of INI-43 to TNFAIP is greater than its affinity for other proteins, such as p65 or IKKβ, which may explain the selectivity of this drug towards tumor cells
