GLPG0634 is a potent inhibitor of the enzyme toll-like receptor (TLR) 4, and it has been shown to show anti-inflammatory activity in vitro. GLPG0634 is currently being investigated for its potential therapeutic effects in bowel disease and arthritis. GLPG0634 binds to TLR4, which is located on the surface of cells, inhibiting the activation of these cells by bacterial products or other agents that stimulate TLRs. This inhibition may be due to the suppression of cytokine production from immune cells such as monocytes and macrophages. The drug also inhibits pro-inflammatory responses mediated by Toll-like receptor 2 signalling pathways, such as those induced by lipopolysaccharide (LPS). GLPG0634 has been found to be effective against LPS-induced inflammation in vivo, with a potency comparable to that seen in vitro.
