- 2-Amino-4-(chloromethyl)thiazole
2-Amino-4-chloromethylthiazole (2ACMT) has been shown to be an effective antifilarial agent in pyridinium, acanthocheilonema, and refluxed mice. 2ACMT is a nucleophilic compound that binds to the active site of the enzyme DNA gyrase, inhibiting DNA synthesis. This drug also has antibacterial activity, which may be due to its ability to inhibit protein synthesis in bacteria.
The drug was found to be toxic at high doses in toxicity studies on mice. It was also found to cause a significant decrease in white blood cells when administered orally or intravenously at doses of 200 mg/kg or higher. 2ACMT is not absorbed into the bloodstream after oral administration and does not cross the blood-brain barrier after intravenous administration. However, it is possible for this drug to affect the central nervous system by crossing the blood-brain barrier through other routes of exposure
