5-Methoxybenzofuroxan is an antitubercular drug that belongs to the class of nitrofurans. It is a prodrug that is hydrolyzed in vivo to benzoquinone and 5-methoxybenzofuran. The biological properties of 5-methoxybenzofuroxan are similar to those of nitrosoureas, with the exception that it inhibits bacterial growth by binding to DNA rather than RNA. This binding prevents transcription and replication. Nitrosoureas have been shown to be active against Mycobacterium tuberculosis, but not against other species such as Mycobacterium avium complex or Mycobacterium leprae. These compounds are also toxic to mammalian cells. The toxicity may be due to their ability to induce apoptosis in a number of cell types, including human erythrocytes and human bronchial epithelial cells (v79).
