Description:

2,3-Dihydro-6-isothiocyanato-1,4-phthalazinedione (DITC) is a trivalent metal chelate that reacts with hydrogen peroxide to produce free radicals. It can be used as a reagent to detect biogenic amines in urine samples. The sensitivity of DITC is increased when derivatization is performed before the reaction with hydrogen peroxide. Derivatization is performed by reacting the sample with an organic molecule containing a reactive group such as hydrazine, carbonyl chloride, or diazomethane. DITC reacts with these molecules and produces hydroxyl radicals in the presence of hydrogen peroxide. Hydroxyl radicals react with the amine groups of biogenic amines such as histamine and tyramine to form adducts that are detectable by spectrophotometry at 412 nm.
