2-Pyridin-3-ylethanamine dihydrobromide is an isothiuronium salt that has been found to be a potent inhibitor of thyroxine (T4) deiodination. It has been shown to inhibit the enzyme with an IC50 of 2.5 μM at pH 7.2 and 10 μM at pH 4.0. The affinity of the ligand for the enzyme is dependent on pH, with a higher affinity being observed at lower pH values. A possible mechanism for inhibition involves the covalent binding of the ligand to the active site cysteine residues in the enzyme, which can be demonstrated by titration experiments where increasing concentrations of substrate lead to decreasing absorbance spectra and increased potency of inhibition.
