Product Information
NNC-0640 is a novel incretin hormone receptor binding agent. It has been shown to be an allosteric modulator of the GLP-1R, and it has been shown to promote insulin secretion in diabetic patients. NNC-0640 binds to the GLP-1R and enhances the affinity of the receptor for its ligand, GLP-1. Binding of this drug to the GLP-1R leads to increased activation of adenylyl cyclase (AC), which results in increased intracellular levels of cyclic AMP (cAMP). This increase in cAMP activates protein kinase A (PKA) by phosphorylation, which then initiates signaling pathways that lead to insulin secretion. The allosteric modulation of NNC-0640 is thought to be due to a conformational change in the receptor caused by binding with this drug.
