4-(2-Methoxyphenyl)piperidine is a piperidine compound that has shown to be a potent inhibitor of histone deacetylase 1 (HDAC1) and 2 (HDAC2). It also inhibits platelet activation, which is mediated by the P2Y1 receptor. 4-(2-Methoxyphenyl)piperidine is a competitive inhibitor of HDACs, binding to the active site of these enzymes. It has been shown to reduce platelet aggregation in vitro. The structure of 4-(2-Methoxyphenyl)piperidine was optimized for binding affinity and potency at the Hdac1 and Hdac2 enzyme targets. A docking study was conducted on the crystal structure of HDAC1, followed by molecular dynamics simulations to investigate its interactions with this enzyme. This led to the identification of two novel inhibitors with improved binding affinity and potency against HDACs 1 and 2.
