Description:

(2S,5R)-5-Phenylpyrrolidine-2-carboxylic acid is a potent inhibitor of bacterial endopeptidase. It irreversibly inhibits the enzyme by binding to the active site and inhibiting peptide cleavage. The inhibition is due to an acrylonitrile group in the molecule that binds to subsites on the enzyme's surface. The inhibition of this enzyme prevents angiotensin I from being converted into angiotensin II, which is a potent vasoconstrictor. (2S,5R)-5-Phenylpyrrolidine-2-carboxylic acid also inhibits thermolysin, a proteolytic enzyme found in many organisms including staphylococci and pyrrolidonyl esterases found in Mycobacterium tuberculosis. This drug has been shown to be effective against methicillin resistant Staphylococcus aureus (
