Tipiracil is a group P2 epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor that has been shown to have a number of physiological effects in various model organisms. Tipiracil has been shown to be effective against inflammatory bowel disease and colorectal adenocarcinoma, and it can inhibit the proliferation of skin cancer cells in mice. Tipiracil binds to EGFR with high affinity, inhibiting its activation by binding to the ATP-binding site on the tyrosine kinase domain. This prevents autophosphorylation and activation of downstream signaling pathways such as MAPKs, leading to inhibition of cell proliferation. Tipiracil also inhibits DNA synthesis and protein synthesis in cells.
