2-Hydroxyphenylacetic acid (2OH-PA) is a metabolite of benzoic acid, a toxin found in certain plants. It has been shown to interact with cellular kinases and inhibit their activity. 2OH-PA interacts with the ATP binding site of the kinase domain by forming a ternary complex with the kinase and ATP. The benzyl group on 2OH-PA tethers it to the enzyme and prevents it from diffusing away, while at high concentrations it can also induce conformational changes in the protein that result in an inhibition of kinase activity. In tumor tissue, 2OH-PA can upregulate p21 expression, which may be due to its ability to inhibit mitotic checkpoint proteins such as Cyclin B1.
