1-(3-tert-Butylphenyl)methanamine is a low nanomolar x-ray crystallographic inhibitor of the toluene hydroxylase, which is an enzyme involved in the conversion of toluene to hydroxyethylamine. It has been shown to have a macrocyclic structure and isostere. 1-(3-tert-Butylphenyl)methanamine inhibits the enzyme by binding to its active site and blocking access to substrate. 1-(3-tert-Butylphenyl)methanamine has also been shown to inhibit the activity of other enzymes, including cytochrome P450s, as well as cell growth in culture. The drug has been shown to be selective for cells that contain the target enzyme and can be used in cell-based assays and enzymatic assays.
