Chlorothricin is an antibiotic that inhibits the growth of bacteria by binding to the 50S ribosomal subunit. It binds to the benzodiazepine binding site in protein and has potent antitumor activity. Chlorothricin has been shown to inhibit protein synthesis in mammalian tissue, which may be due to its ability to bind with malonic acid or hydroxyl groups. Chlorothricin is synthesized by a complex set of reactions involving enzymatic modification of chlorotetracenyl diphosphate (CTDP). The CTDP can be converted into chlorothricin either by hydrolysis with esterases, oxidation with cytochrome P450 enzymes, reduction by glutathione reductase, or conjugation with glucuronic acid.
