Pioglitazone is a drug that belongs to the class of thiazolidinediones. It is a synthetic compound, which is metabolized in the liver and excreted by the kidneys. Pioglitazone-d4 has been used as an endogenous substance marker for pharmacokinetic studies. The plasma concentration-time curve of pioglitazone-d4 in humans was found to be very similar to that for pioglitazone, with retention time at about 2 hours. The elimination half-life of pioglitazone-d4 was found to be about 7 hours, which is significantly longer than that of pioglitazone. Pioglitazone-d4 provides a more stable chromatographic peak than does pioglitazone and can be used for matrix effect analysis.
