Benzoic acid, 3-amino-5-nitro-, ethyl ester (BA3NE) is a potent inhibitor of the PI3K/AKT pathway. BA3NE has been shown to inhibit proliferation and induce apoptosis in a number of cancer cell lines. It also inhibits the growth of human leukemia cells by inducing cell cycle arrest at G2/M phase. The mechanism of action is not known, but it may be due to inhibition of phosphorylation of Akt or downstream targets such as glycogen synthase kinase 3β (GSK3β). BA3NE was found to be more efficient than SB239063, another PI3K/AKT pathway inhibitor, with a Ki value that is about 100 times lower for both the ATP and ATPγS binding site.
