PKC is activated by diacylglycerol, which is synthesized from phosphatidylinositol 4,5-bisphosphate. The inhibition of PKC prevents the activation of various downstream events including ion channels, the transcription factor NF-κB, and the production of proinflammatory cytokines. W 212393 binds to PKC with high affinity and specificity. It also inhibits other members of the family of protein kinases, including protein kinase A and glycogen synthase kinase 3β.
