Foropafant is a small molecule that inhibits transcription by binding to the polymerase-binding site of the DNA strand. Foropafant was developed as a potential treatment for cancer, but has shown effects on other cell types. It binds to the basic fibroblast growth factor receptor (bFGF-R) and this binding leads to changes in cellular physiology. Foropafant also blocks protein synthesis at the level of initiation by inhibiting dna binding activity and blocking protein synthesis. In vivo studies have shown that foropafant penetrates into tumor tissue and inhibits tumor growth, which may be due to its ability to inhibit transcription or protein synthesis in cells with bFGF-R receptors.
